C1.Ori

C1 is a minimal autonomously replicating sequence (ARS) and centromeric sequence derived from the number 7 chromosome of Kluyveromyces marxianus strain DMKU3-1042 [1]. This enables the in vitro construction of cloning and expression systems for K. marxianus with high flexibility.


Usage and Biology

The ARS contains the yeast genome’s origin of replication, including four regions: A, B1, B2, B3. It is A-T rich thus the hydrogen bonding within the ARS is easily disrupted by replicative proteins[1]. Origin recognition protein complex is bound to it throughout the cell cycle, allowing the access of replicative proteins.
